Research and analytical settings increasingly require documentation of peptide origin and chain of custody. Certificate of analysis documents typically report purity by chromatographic area, mass confirmation, appearance, and residual solvent or counterion content. Independent verification by an accredited laboratory is common when a material will be used in a regulated study. Open questions remain about how well compendial methods transfer between laboratories, and about which impurity thresholds are meaningful for materials not intended for clinical use.
Peptide-based pharmaceutical products such as tirzepatide require controlled temperature management to preserve structural integrity. Manufacturer labeling generally specifies refrigeration at 2 to 8 degrees Celsius before first use, with protection from light and freezing. Exposure to repeated temperature cycling can promote aggregation or deamidation, which alters the analytical profile even when the visible solution appears unchanged. Once a product is in use, the permitted storage window and temperature range are defined by the specific labeled presentation rather than by general peptide rules.
Identity and purity assessment of tirzepatide relies primarily on reversed-phase high-performance liquid chromatography coupled with ultraviolet detection. Mass spectrometry, often in electrospray ionization mode, confirms the molecular mass and detects sequence-related impurities. Peptide mapping after enzymatic digestion provides residue-level confirmation of the backbone. Each method addresses a different question: chromatography for purity and related substances, mass measurement for identity, and mapping for sequence fidelity. No single technique covers all three.
Development of tirzepatide took place under a research program that sought to test whether simultaneous engagement of two incretin receptors would produce greater metabolic effects than single-receptor agonism. Clinical trials were organized into the SURPASS series for type 2 diabetes and the SURMOUNT series for obesity and weight management. Regulatory clearance for type 2 diabetes came in 2022 in the United States, followed by approval for chronic weight management in 2023. The trial programs reported reductions in glycated hemoglobin and body weight relative to comparators, though long-term cardiovascular and durability data continue to accumulate.
The peptide backbone contains 39 amino acids and includes alpha-aminoisobutyric acid residues, which are not among the standard proteinogenic set. A C20 fatty diacid moiety is attached through a linker, allowing the compound to bind serum albumin and extend its circulation time. This albumin binding is the main reason the molecule supports once-weekly administration rather than more frequent dosing. The measured molecular mass is approximately 4,813 daltons, placing it firmly in the peptide rather than small-molecule class.

Lyophilized material is generally held at -20 degrees Celsius or lower, desiccated and protected from light, where it remains stable for extended periods. Reconstituted or ready-to-use solution is usually kept at 2 to 8 degrees Celsius with minimal agitation. Repeated freeze-thaw cycles should be avoided because they promote aggregation and reduce the soluble monomer fraction. Shipment of frozen solid commonly uses dry ice, while refrigerated liquid moves with validated cold packs. Stability beyond documented periods is not established.
Characterization of the peptide relies on reversed-phase high-performance liquid chromatography for purity and related-substance profiling, with ultraviolet detection near 214 nanometers. Mass spectrometry confirms molecular mass and reveals modifications such as oxidation or deamidation. Peptide mapping after enzymatic digestion verifies the amino acid sequence, while amino acid analysis supplies compositional data. Circular dichroism and infrared spectroscopy are used to assess secondary structure, particularly the alpha-helical content that influences aggregation behavior in solution.
After subcutaneous injection, absorption is gradual, and peak plasma levels are generally reached within one to three days. Albumin binding extends the apparent half-life to roughly five days, which supports a weekly administration schedule. Metabolism proceeds mainly through proteolytic cleavage of the peptide backbone and beta-oxidation of the fatty acid chain, rather than through cytochrome P450 pathways. Eliminated fragments are largely recycled through general protein turnover, and excretion of intact drug in urine is minimal. These properties distinguish the molecule from short-acting incretin mimetics.
Tirzepatide is a synthetic peptide of 39 amino acids engineered from the native glucose-dependent insulinotropic polypeptide sequence. Its structure incorporates several non-natural residues and a C-terminal segment derived from glucagon-like peptide-1, together with a C20 fatty diacid moiety attached through a linker. The lipophilic side chain promotes binding to serum albumin, which slows renal clearance after administration. The compound is classified as a dual incretin receptor agonist and is supplied as a lyophilized powder for reconstitution or as a preformulated solution, depending on the presentation.
== Background == Robert Karriem was born Robert Harris on August 3, 1888, to parents Alec and Lulu in Mississippi. On July 3, 1929, he moved to Detroit, Michigan. He had a wife, Bertha. James Smith was born on December 25, 1892, in Atlanta. In 1930, a man named W. D. Fard came to Detroit, ultimately founding a group called the Allah Temple of Islam. Among Fard's practices was to give new, Islamic names to members, replacing their inherited 'slave names'. Harris was given the new surname "Karriem".
== Epidemiology == Kwashiorkor is rare in high-income countries. It is mostly observed in low-income and middle-income nations and regions such as Southeast Asia, Central America, Congo, Ethiopia, Puerto Rico, Jamaica, South Africa, and Uganda, where poverty is prominent. Occurrences of severe malnutrition also tend to trend higher under conditions of food insecurity, higher prevalence of infectious diseases, lack of access to appropriate care, and poor living situations with inadequate sanitation. Communities experiencing famine are affected the most, especially during the rainy season. Prevalence varies, but it affects children of either sex commonly under five years old. "Globally, kwashiorkor indirected accounted for 53% of deaths among children under five between 2000 and 2003 when associated with other common childhood diseases like acute respiratory infections, malaria, measles, HIV/AIDS and other causes of perinatal deaths." When compared to marasmus in developing countries, kwashiorkor typically has a lower prevalence, "0.2%-1.6% for kwashiorkor and 1.2%-6.8% for marasmus." Factors such as "diet, geographical locations, climate, and aflatoxin exposure" have been invoked as potential causes for observed differences in the prevalence of kwashiorkor and marasmus. In general, in areas where Severe Acute Malnutrition (SAM) is prevalent, marasmus is more often the dominant SAM condition. However, in certain areas, kwashiorkor may be more common than marasmus.
Keratolytic () therapy is a type of medical treatment to remove warts, calluses and other lesions in which the epidermis produces excess skin. In this therapy, acidic topical medicines, such as Whitfield's ointment or Jessner's solution, are applied to the lesion in order to thin the skin on and around it. This therapy causes the outer layer of the skin to loosen and shed. Keratolytics can also be used to soften keratin, a major component of the skin. This serves to improve the skin's moisture binding capacity, which is beneficial in the treatment of dry skin. Such agents (keratolytics) include alkalis (by swelling and hydrolysis of skin), salicylic acid, urea, lactic acid, allantoin, glycolic acid, and trichloroacetic acid. While cytostatic agents such as zinc pyrithione are first line, keratolytics (salicylic acid and sulfur) can also be used in the treatment of dandruff and seborrheic dermatitis. Sulfur and salicylic acid can also be used to effectively treat acne and cradle cap in some patients. Resorcinol is another keratolytic that is usually combined with sulfur. Urea acts as a keratolytic due to its hygroscopic property.
In surgical praxis, after having emplaced the empty breast implants to the implant pockets, the plastic surgeon then filled each device with saline solution, and, because the required insertion-incisions are short and small, the resultant incision-scars will be smaller and shorter than the surgical scars usual to the long incisions required for inserting pre-filled, silicone-gel implants. When compared to the results achieved with a silicone-gel breast implant, the saline implant can yield acceptable results, of increased breast-size, smoother hemisphere-contour, and realistic texture; yet, it is likelier to cause cosmetic problems, such as the rippling and the wrinkling of the breast-envelope skin, accelerated lower breast pole stretch, and technical problems, such as the presence of the implant being noticeable to the eye and to the touch. The occurrence of such cosmetic problems is likelier in the case of the woman with very little breast tissue, and in the case of the woman who requires post-mastectomy breast reconstruction; thus, the silicone-gel implant is the technically superior prosthetic device for breast augmentation, and for breast reconstruction. In the case of the woman with much breast tissue, for whom sub-muscular emplacement is the recommended surgical approach, saline breast implants can produce an aesthetic result much like that afforded by silicone breast implants. Ultrasound examination and outcome studies have revealed that saline and silicone breast implants look and feel similar.

== Lawsuits == In November 2016, three men filed a class-action lawsuit against Chipotle alleging that a burrito was listed as containing only 300 calories, when in fact it contained more. They are seeking "unspecified damages and for an injunction against the company to prevent it from posting what it calls misleading information on its food". In January 2020, the company was fined $1.3 million by the state of Massachusetts for 13,000 child labor violations.
2C-B was legitimately marketed and sold as an over-the-counter sexual enhancer under brand names like Erox in several European countries such as Germany in the 1980s and early 1990s. It was manufactured by the German pharmaceutical company Drittewelle and was sold in adult stores, smart shops, and some nightclubs. In addition, 2C-B was sold in Dutch smart shops as an ecstasy-like legal high under names like Nexus. 2C-B was first encountered as a novel recreational designer drug in the United States in 1985. It was advertised and used as an MDMA substitute after MDMA was made illegal around this time. 2C-B has been said to have been legally sold in Southern Africa from 1993 to 1996 and used as an entheogen by the Sangoma, Nyanga, and Amagqirha people in place of their traditional plants; they refer to the chemical as Ubulawu Nomathotholo, which roughly translates to "Medicine of the Singing Ancestors". The drug became a controlled substance in the United States in 1994. It also became a controlled substance in most other countries in the mid-1990s. In addition, 2C-B was placed in Schedule II of the United Nations Convention on Psychotropic Substances and hence became an internationally controlled substance in 2001. Following 2C-B's restriction, many other 2C psychedelics, such as 2C-E and 2C-I, emerged as designer drugs. Nonetheless, 2C-B is the most popular of the 2C psychedelics. Subsequent to their emergence, numerous other 2C drugs besides 2C-B have also been made controlled substances throughout the world.
Albert Lester Lehninger (February 17, 1917 – March 4, 1986) was an American chemist in the field of bioenergetics. He made fundamental contributions to the current understanding of metabolism at a molecular level. In 1948, he discovered, with Eugene P. Kennedy, that mitochondria are the site of oxidative phosphorylation in eukaryotes, which ushered in the modern study of energy transduction. He is the author of a number of classic texts, including Biochemistry, The Mitochondrion, Bioenergetics and, most notably, his series Principles of Biochemistry. This last is a widely used text for introductory biochemistry courses at the college and university levels. Lehninger was born in Bridgeport, Connecticut, US. He earned his BA in English from Wesleyan University (1939) and went on to earn both his MA (1940) and PhD (1942) at the University of Wisconsin–Madison. His doctoral research involved the metabolism of acetoacetate and fatty acid oxidation by liver cells.
After several months of work, Fermi's group had tentatively determined that lighter elements would disperse the energy of the captured neutron by emitting a proton or alpha particle and heavier elements would generally accomplish the same by emitting a gamma ray. This latter behavior would later result in the beta decay of a neutron into a proton, thus moving the resulting isotope one place up the periodic table. When Fermi's team bombarded uranium, they observed this behavior as well, which strongly suggested that the resulting isotope had an atomic number of 93. Fermi was initially reluctant to publicize such a claim, but after his team observed several unknown half-lives in the uranium bombardment products that did not match those of any known isotope, he published a paper entitled Possible Production of Elements of Atomic Number Higher than 92 in June 1934. For element 93, he proposed the name ausenium (atomic symbol Ao) after the Greek name Ausonia for Italy. Several theoretical objections to the claims of Fermi's paper were quickly raised; in particular, the exact process that took place when an atom captured a neutron was not well understood at the time. This and Fermi's accidental discovery three months later that nuclear reactions could be induced by slow neutrons cast further doubt in the minds of many scientists, notably Aristid von Grosse and Ida Noddack, that the experiment was creating element 93.
The vorticity equation of fluid dynamics describes the evolution of the vorticity ω of a particle of a fluid as it moves with its flow; that is, the local rotation of the fluid (in terms of vector calculus this is the curl of the flow velocity). The governing equation is:where D/Dt is the material derivative operator, u is the flow velocity, ρ is the local fluid density, p is the local pressure, τ is the viscous stress tensor and B represents the sum of the external body forces. The first source term on the right hand side represents vortex stretching. The equation is valid in the absence of any concentrated torques and line forces for a compressible, Newtonian fluid. In the case of incompressible flow (i.e., low Mach number) and isotropic fluids, with conservative body forces, the equation simplifies to the vorticity transport equation:

(131.5 kg) Ladd, the tallest player in professional football, was All-AFL three times (1961, 1964-65) and an AFL All-Star from 1962 to 1965. Nery and Hudson played with Faison and Ladd in 1961 and 1962, Hudson being named an All-Star in 1961, and Nery selected All-AFL in 1962. Faison and Ladd were later joined by right defensive end Bob Petrich (1963-65), defensive tackle George Gross (1963-65), and Henry Schmidt who had been a reserve lineman and then had starter roles at tackle and end (1961-65). At the time Gross and Ladd were two of the largest and strongest men in professional football. From 1961 to 1965, the Chargers played in four of the five AFL Championship games, winning the 1963 AFL Championship Game, 51–10, over the Boston Patriots.
=== Aviation === In 2016, researchers developed a prototype de-icing system that incorporated unzipped carbon nanotube graphene nanoribbons in an epoxy/graphene composite. In laboratory tests, the leading edge of a helicopter rotor blade was coated with the composite, covered by a protective metal sleeve. Applying an electrical current heated the composite to over 200 °F (93 °C), melting a 1 cm (0.4 in)-thick ice layer with ambient temperatures of a -4 °F (-20 °C).

== Function == Glutathione synthetase is important for a variety of biological functions in multiple organisms. In Arabidopsis thaliana, low levels of glutathione synthetase have resulted in increased vulnerability to stressors such as heavy metals, toxic organic chemicals, and oxidative stress. The presence of a thiol functional group allows its product GSH to serve both as an effective oxidizing and reducing agent in numerous biological scenarios. Thiols can easily accept a pair of electrons and become oxidized to disulfides, and the disulfides can be readily reduced to regenerate thiols. Additionally, the thiol side chain of cysteines serve as potent nucleophiles and react with oxidants and electrophilic species that would otherwise cause damage to the cell. Interactions with certain metals also stabilize thiolate intermediates. In humans, glutathione synthetase functions in a similar manner. Its product GSH participates in cellular pathways involved in homeostasis and cellular maintenance. For instance, glutathione peroxidases catalyze the oxidation of GSH to glutathione disulfide (GSSG) by reducing free radicals and reactive oxygen species such as hydrogen peroxide. Glutathione S-transferase uses GSH to clean up various metabolites, xenobiotics, and electrophiles to mercapturates for excretion. Because of its antioxidant role, GSS mostly produce GSH inside the cytoplasm of liver cells and imported to mitochondria where detoxification occurs. GSH is also essential for the activation of the immune system to generate robust defense mechanisms against invading pathogens.
== Use == Areas of increased pigmentation such as moles may be depigmented to match the surrounding skin. Effective agents for specific areas include corticosteroids, tretinoin, and hydroquinone. These agents are not allowed in cosmetics in Europe due to concerns about side effects. Attempts to whiten large areas of skin may also be carried out by certain cultures. This may be done for reasons of appearance, politics, or economics. Skin whiteners can help achieve lighter skin tones, but many of them contain harmful ingredients, such as the steroid clobetasol propionate, inorganic mercury (mercuric chloride or amalgamated mercury), glutathione (an antioxidant traditionally used in cancer treatment), and the organic compound hydroquinone. Skin lighteners' main health risks are linked to (i) The overuse of topical clobetasol, which can cause systemic steroid effects from daily usage, especially on broad skin regions; and (ii) concealed mercury content, which can lead to mercury poisoning depending on individual susceptibility. Many skin whiteners contain a toxic form of mercury as the active ingredient. Their use, therefore, may harm a person's health and is illegal in many countries.
