This is a working overview of albumin binding, written for readers who want more than a one-paragraph summary but less than a textbook.

Clinical development proceeded through large phase 3 programmes in type 2 diabetes and in obesity or overweight with at least one weight-related comorbidity. Regulatory approvals followed in several jurisdictions for both indications. Weekly subcutaneous dosing reflects an elimination half-life of roughly five days. Open questions include the durability of metabolic effects after treatment stops, long-term cardiovascular and hepatic outcomes beyond completed trials, and whether the dual mechanism confers benefits independent of total receptor occupancy. Published literature continues to expand on these points. Substantial uncertainty remains about interindividual variability in response.
Tirzepatide is a synthetic linear peptide of 39 amino acids that acts as a dual agonist at the glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) receptors. Its sequence derives from native GIP but incorporates non-natural residues and a C20 fatty diacid moiety linked to a lysine side chain. The lipophilic chain promotes albumin binding, which slows renal clearance and extends circulation time. The unmodified peptide has a molecular formula of C225H348N48O68 and a molecular mass near 4,813 daltons.
Receptor activation by tirzepatide raises intracellular cyclic AMP through Gs-coupled signalling at both targets. At the GLP-1 receptor the downstream effect includes glucose-dependent insulin release, suppressed glucagon secretion, delayed gastric emptying, and reduced appetite signalling in the hypothalamus. GIP receptor engagement adds insulinotropic activity and appears to influence lipid handling in adipose tissue. Because both receptors are stimulated at the same time, the pharmacological profile differs from that of selective GLP-1 receptor agonists, and the relative contribution of each arm remains an area of active investigation.
Structural work on the molecule centers on a C20 fatty diacid moiety attached through a linker to the peptide backbone. This side chain promotes reversible binding to serum albumin, which slows renal clearance and supports a prolonged action profile. The peptide backbone incorporates aminoisobutyric acid substitutions that limit recognition by digestive enzymes. Together these modifications produce a molecule that is stable enough for subcutaneous delivery but still dependent on careful manufacturing control. Analytical characterization of the active pharmaceutical ingredient typically follows the conventions used for other synthetic peptides.
Tirzepatide is a synthetic peptide composed of 39 amino acids. It acts as a dual agonist at two incretin receptors, the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. The molecule was designed by modifying the native sequence of glucose-dependent insulinotropic polypeptide to improve metabolic stability and extend its circulation time. Its structure includes several non-natural amino acid residues and a fatty acid side chain. These features distinguish it from earlier single-receptor incretin analogs studied in the same period.

At the receptor level, tirzepatide activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. Both belong to the class B family of G protein-coupled receptors and signal largely through cyclic AMP accumulation. The compound binds the two receptors with differing affinity, and the pattern of signaling at each site is described in the literature as biased rather than simply proportional to occupancy. Tissues carrying these receptors include pancreatic islets, adipose tissue, the central nervous system, and the gastrointestinal tract. The relative weight of each receptor population in producing metabolic effects continues to be studied.
Published work supports the view that engaging two incretin receptors produces changes in glucose handling and body weight larger than those seen with single-receptor activation. Why that difference arises is not fully settled. Open questions include how much of the observed weight effect depends on central versus peripheral signaling, and whether the two receptors form interacting complexes. Most reported findings come from controlled trials and animal models, and translation between species is imperfect. Further research is expected to refine these points over time.
Tirzepatide is a synthetic peptide built from 39 amino acid residues. Its sequence is related to human glucose-dependent insulinotropic polypeptide, with modifications that include a C-terminal extension and a C20 fatty diacid joined through a linker. Those changes raise the molecule's affinity for serum albumin, which slows renal filtration and lengthens the time it stays in circulation. The free base has an average molecular mass near 4813.5 daltons. The compound is made by solid-phase peptide synthesis followed by chromatographic purification.
=== Cancer === While serum low 25-hydroxyvitamin D status has been associated with a higher risk of cancer in observational studies, the general conclusion is that there is insufficient evidence for an effect of vitamin D supplementation on the risk of cancer, although there is some evidence for reduction in cancer mortality.
That said, Jung's concept of individuation—that is the integration of the archaic unconscious with consciousness—seems extremely pertinent to modern psychedelic research." The author also uses work in evolutionary and psychedelic neuroscience, and specifically the latter's ability to make manifest ancient subcortical brain systems, to illuminate Jung's concept of an archaic collective unconscious that evolved before the ego complex and the uniquely human default mode network.
After rail services were extended to Dover in 1844, the Western Docks were used as a terminal for the Golden Arrow and other cross-channel train services with its own railway station, Dover Marine, later renamed Dover Western Docks. At Dover Marine station an estimated 5 million troops departed for the trenches of World War I and nearly 1.5 million wounded soldiers returned. In 1920 the remains of the Unknown Warrior were landed prior to transportation to London and a ceremonial interment at Westminster Abbey. Dover Marine railway station closed in 1994, later to be redeveloped into Cruise Terminal One. The Western Docks area was also used from 1968 to the early 2000s for a cross-channel hovercraft service run by Hoverspeed. Hoverspeed also ran catamaran services until being declared bankrupt in 2005. Another catamaran service was run from 2004 until November 2008 by the single ship of SpeedFerries, SpeedOne, with up to five services daily to Boulogne-sur-Mer. The hoverport has now been demolished and redeveloped as a cargo handling facility.
However, there is significant charge transfer between the two materials and, in some cases, hybridization between the d-orbitals of the substrate atoms and π orbitals of graphene, which significantly alter the electronic structure compared to that of free-standing graphene. Boehm et al. coined the term "graphene" for the hypothetical single-layer structure in 1986. The term was used again in 1987 to describe single sheets of graphite as a constituent of graphite intercalation compounds, which can be seen as crystalline salts of the intercalant and graphene. It was also used in the descriptions of carbon nanotubes by R. Saito and Mildred and Gene Dresselhaus in 1992, and in the description of polycyclic aromatic hydrocarbons in 2000 by S. Wang and others. Efforts to make thin films of graphite by mechanical exfoliation started in 1990. Initial attempts employed exfoliation techniques similar to the drawing method. Multilayer samples down to 10 nm in thickness were obtained. In 2002, Robert B. Rutherford and Richard L. Dudman filed for a patent in the US on a method to produce graphene by repeatedly peeling off layers from a graphite flake adhered to a substrate, achieving a graphite thickness of 0.00001 inches (0.00025 millimetres). The key to success was the ability to quickly and efficiently identify graphene flakes on the substrate using optical microscopy, which provided a small but visible contrast between the graphene and the substrate. Another U.S. patent was filed in the same year by Bor Z. Jang and Wen C.

== Interactions == A number of drugs add to the low blood pressure caused by nitrovasodilators: for example, other vasodilators, antihypertensive drugs, tricyclic antidepressants, antipsychotics, general anaesthetics, as well as ethanol. Combination with PDE5 inhibitors, including sildenafil (Viagra), is contraindicated because potentially life-threatening hypotension may occur. Nitrates increase the bioavailability of dihydroergotamine (DHE). High DHE levels may result in coronary spasms in patients with coronary disease. This interaction is not described for non-nitrate nitrovasodilators.
23793Np + 4820Ca → 285113* → 282113 + 3 n Two atoms of 282113 were detected. The aim of this experiment had been to synthesise the isotopes 281113 and 282113 that would fill in the gap between isotopes produced via hot fusion (283113 and 284113) and cold fusion (278113). After five alpha decays, these nuclides would reach known isotopes of lawrencium, assuming that the decay chains were not terminated prematurely by spontaneous fission. The first decay chain ended in fission after four alpha decays, presumably originating from 266Db or its electron-capture daughter 266Rf. Spontaneous fission was not observed in the second chain even after four alpha decays. A fifth alpha decay in each chain could have been missed, since 266Db can theoretically undergo alpha decay, in which case the first decay chain would have ended at the known 262Lr or 262No and the second might have continued to the known long-lived 258Md, which has a half-life of 51.5 days, longer than the duration of the experiment: this would explain the lack of a spontaneous fission event in this chain. In the absence of direct detection of the long-lived alpha decays, these interpretations remain unconfirmed, and there is still no known link between any superheavy nuclides produced by hot fusion and the well-known main body of the chart of nuclides.
== Signs and symptoms == The hallmark of polymyositis is weakness and/or loss of muscle mass in the proximal musculature, as well as flexion of the neck and torso. These symptoms can be associated with marked pain in these areas as well. The hip extensors are often severely affected, leading to particular difficulty in climbing stairs and rising from a seated position. The skin involvement of dermatomyositis is absent in polymyositis. Dysphagia (difficulty swallowing) or other problems with esophageal motility occur in as many as 1/3 of patients. Low grade fever and enlarged lymph nodes may be present. Foot drop in one or both feet can be a symptom of advanced polymyositis and inclusion body myositis. The systemic involvement of polymyositis includes interstitial lung disease (ILD) and heart disease, such as heart failure and conduction abnormalities. Polymyositis tends to become evident in adulthood, presenting with bilateral proximal muscle weakness often noted in the upper legs due to early fatigue while walking. Sometimes the weakness presents itself as an inability to rise from a seated position without help or an inability to raise one's arms above one's head. The weakness is generally progressive, accompanied by lymphocytic inflammation (mainly cytotoxic T cells).
==== Insects ==== In insects, a system involving Malpighian tubules is used to excrete metabolic waste. Metabolic waste diffuses or is actively transported into the tubule, which transports the wastes to the intestines. The metabolic waste is then released from the body along with fecal matter. The excreted material may be called ejecta. In pathology the word ejecta is more commonly used.

In Australia, two distinctly different coloured sodas exist, red and brown, both usually called creaming soda, although some brands such as Bickford's, use the term creamy soda. Almost all varieties are predominantly vanilla based, but red or pink varieties introduce raspberry flavoring. Another local variant produced by Golden Circle is vanilla and fruit-flavored, and coloured yellow to distinguish it from existing brands. More traditional brown varieties are also available, but less common. Brands include Kirks' Sno Drop (only available in South Australia, Victoria, and the Northern Territory), Tarax, River Port, Hartz, Saxby's, Bert's Snowette (the original recipe of Shelley's Snowcap (Snowcap Champagne) before the line was acquired) and Schweppes, which also produce a red variety as part of its "Traditionals" range. Bundaberg Burgundee creaming soda is based on red grapes and is alternately made by other producers under the name portello. The term "creaming soda" is used to refer to the drink itself, whereas the combination of soda and ice-cream is called a spider.
== Pollution == C. brunneus are used as a bioindicators for heavy metal pollution. They are commonly found living in habitats that are polluted with heavy metals such as Szopienice and Olkusz in Poland. Some sites have heavy metal concentrations as high as 124.3±15.9 mg•kg-1. Individuals can have heavy metal concentrations as larger as 21.25 mg•kg-1. Exposure to heavy metal concentrations alters catalytic ability of enzymes. Individuals from heavily polluted sites have increased glutathione concentrations and decreased glutathione S-transferase activity. In the lab, individuals exposed to zinc during diapause have lower glutathione concentrations. Dimethoate exposure enhances the effect of heavy metal exposure decreasing glutathione concentrations and reducing acetylcholinesterase activity by almost 50%. Exposure to dimethoate also decreases glutathione peroxidase, glutathione reductase, and carboxylesterases activity. Because C. brunneus in non polluted reference sites do not experience the same decrease in enzyme activities, researchers have suggested that the decreased enzyme activities can be contributed to the tradeoff associated with adapting to living in heavily polluted habitats. Individuals are forced to allocate more energy towards neutralizing harmful effects of heavy metals instead of allocating the energy to growth and development.
Phosphorylation changes the conformation of an enzyme to a more active or inactive way (e.g. regulation of glycogen phosphorylase). Each phosphate group contains two negative charges, so the addition of this group can cause an important change in the conformation of the enzyme. The phosphate can attract positively charged amino acids or create repulsive interactions with negatively charged amino acids. These interactions can change the conformation and the function of the enzyme. When a phosphatase enzyme removes the phosphate groups, this enzyme returns to its initial conformation. Phosphorylation modifies the affinity of the enzyme to the substrate (e.g. phosphorylation of isocitrate dehydrogenase creates electrostatic repulsion which inhibits the union of the substrate to the active center). Phosphorylation can take place in the active center of the enzyme. It can change the conformation of this active center, so it can recognize the substrate or not. Also, the ionized phosphate can attract some parts of the substrate, which can join to the enzyme. Phosphorylation and dephosphorylation may take place as a result of the response to signals that warn about a change in the cell state. This means that some pathways where regulatory enzymes participate are regulated by phosphorylation after a specific signal: a change in the cell. Some enzymes can be phosphorylated in multiple sites.
=== Campaign methods === Given the high national profile that Sanders maintained since his 2016 campaign, NPR described him as "no longer an underdog" when he announced his 2020 campaign. Using the large email list it built during the 2016 campaign, the 2020 campaign recruited more than one million volunteers within weeks of its launch. It enlisted several former NowThis News employees to produce professional videos for wide social media distribution, live-streamed various forums to its millions of social media followers, and launched a podcast and smartphone app for grassroots organizing.

It is a dual GIP and GLP-1 receptor agonist, frequently grouped with incretin-based peptide therapeutics. It is a peptide rather than a small molecule and is given by subcutaneous injection.
Selective agents engage only the GLP-1 receptor, whereas tirzepatide activates GIP and GLP-1 receptors simultaneously. This difference in receptor coverage is the principal pharmacological distinction emphasised in comparative reviews.
Downstream signalling is partly characterised, but the quantitative contribution of GIP versus GLP-1 receptor activation to metabolic outcomes is not settled. Review articles commonly flag this as an unresolved question rather than a settled finding.
It activates both the glucose-dependent insulinotropic polypeptide receptor and the glucagon-like peptide-1 receptor. This dual activity separates it from agents that act on only one of the two receptors. The relative contribution of each receptor to clinical effects remains an open area of study.
